  • Overview

    Upf1 is a protein that is part of a post-splicing multiprotein complex involved in both mRNA nuclear export and mRNA surveillance. mRNA surveillance detects exported mRNAs with truncated open reading frames and initiates nonsense-mediated mRNA decay (NMD). When translation ends upstream from the last exon-exon junction, this triggers NMD to degrade mRNAs containing premature stop codons. In mammalian cells NMD generally requires that translation terminates upstream of a post-splicing exon junction complex (EJC). The subsequent binding of Upf1 to the EJC triggers Upf1 phosphorylation. Data suggests that phospho-Upf1 interacts directly with eif3 and inhibits the eif3-dependent conversion of 40S ribosome complex to translationally competent 80S ribosome initiation complex to repress continued translation initiation.
