  • Overview

    The p73 gene has been mapped to the short arm of chromosome 1, position 1p36, a region frequently deleted in neuroblastomas. Allelic loss at 1p36 has also been reported in melanoma, breast carcinoma and colon carcinoma. The p73 transcript is differentially spliced to generate 2 mRNAs that code for the 2 isoforms of p73, p73alpha and p73beta. There is significant sequence homology between amino acid residues in the p73 isoforms and the DNA-binding, transactivation and oligomerization domains of p53. p73 activates p21WAF1 in transfected SK-N-AS cells and transfected SAOS2 cells, suggesting a role for p73 as a transcription factor. In similar transfection experiments, p73 was reported to induce apoptosis in SAOS2 and BHK cells.
