Q&As

Does the sample need to be pretreated?

There is no need for pretreatment or purification of samples.

What are the requirements for test samples?

Cell samples should be cultured in choline chloride free medium.

The samples are already frozen in 0.1%SDS, 1%deoxycholate, 1% NP-40, NaCl and 20mM Tris pH7.4. Can these samples be used with this kit nevertheless?

Theoretically, all the components in the customers buffer looks compatible with the assay. This will have to be specifically determined by performing a small pilot scale experiment by preparing one sample in the kits own assay buffer and the buffer and running the assay simultaneously to determine if there are any changes in the reading.

Could it be used to determine the acetylcholine content in sample of Krebs solution obtained by organ baths in which we suspended colonic longitudinal muscle strips?

For tissue I can recommend to homogenise these in assay buffer or lyse them in assay buffer for ten minutes on ice. Centrifuge to remove debris. Lysates can be tested directly.

Can the customer use Tissue culture SN?

Tissue culture SN can be used with this assay.

I want to know if trace amounts of acetylcholine can be detected?

Linear detection range: colorimetric assay 10 to 200 μM, fluorimetric assay 0.4 to 10 μM acetylcholine.

Can this kit be used with rat samples? I want to know how much of homology is their between the human and rat protein. With this information I believe I will feel more confident to use these kits on rat blood.

This kit should work as it does not contain any human specific antibody. The species is irrelevant for this product.

We want to know the amount of acetylcholine in our plasma samples. If the assay measures total choline when acetylcholinesterase is added, how do we know the amount of free choline to ultimately determine acetylcholine?

It will be necessary to set up 2 different sets of wells for measuring the total choline and the free choline in this assay. Therefore, to measure the acetylcholine, it will be necessary to set up duplicate wells for the total choline in the samples and duplicate wells for the free choline in the same samples. I can confirm that the acetylcholine would be the total choline - free choline.

Can this kit be used for an in-cell assay? Can the probe permeabilize the membrane of live cells?

it cannot be used for in-cell assay. It is meant for cell lysates.
