Ask a questionThere is no need for pretreatment or purification of samples.
Cell samples should be cultured in choline chloride free medium.
Theoretically, all the components in the customers buffer looks compatible with the assay. This will have to be specifically determined by performing a small pilot scale experiment by preparing one sample in the kits own assay buffer and the buffer and running the assay simultaneously to determine if there are any changes in the reading.
For tissue I can recommend to homogenise these in assay buffer or lyse them in assay buffer for ten minutes on ice. Centrifuge to remove debris. Lysates can be tested directly.
Tissue culture SN can be used with this assay.
Linear detection range: colorimetric assay 10 to 200 μM, fluorimetric assay 0.4 to 10 μM acetylcholine.
This kit should work as it does not contain any human specific antibody. The species is irrelevant for this product.
It will be necessary to set up 2 different sets of wells for measuring the total choline and the free choline in this assay. Therefore, to measure the acetylcholine, it will be necessary to set up duplicate wells for the total choline in the samples and duplicate wells for the free choline in the same samples. I can confirm that the acetylcholine would be the total choline - free choline.
it cannot be used for in-cell assay. It is meant for cell lysates.
