Description : | Activins, members of the TGF-beta superfamily, are disulfide-linked dimeric proteins originally purified from gonadal fluids as proteins that stimulated pituitary follicle stimulating hormone (FSH) release. Activin proteins have a wide range of biological activities, including mesoderm induction, neural cell differentiation, bone remodeling, hematopoiesis and roles in reproductive physiology. Activin isoforms and other members of the TGF-beta superfamily exert their biological effects by binding to heteromeric complexes of a type I and a type II serine-threonine kinase receptor, both of which are essential for signal transduction.Activins are homodimers or heterodimers of the various beta subunit isoforms, while inhibins are heterodimers of a unique alpha subunit and one of the various beta subunits. Five beta subunits (mammalian beta A, beta B, beta C, beta E and Xenopus beta D) have been cloned to date. The activin/inhibin nomenclature reflects the subunit composition of the proteins: Activin A (beta A - beta A), Activin B (beta B - beta B), Activin AB (beta A - beta B), Inhibin A (alpha - beta A) and Inhibin B (alpha - beta B). |
Source : | CHO cells |
Species : | Human |
Bio-activity : | Measured by its ability to induce hemoglobin expression in K562 cells (Schwall, R.H. et al., 1991, Method Enzymol. 198:340). |
Molecular Mass : | The mature recombinant human Activin AB, generated by proteolytic removal of the propeptides, is a disulfide-linked heterodimer of the mature human Activin βA chain and mature human Activin βB chain. Based on N-terminal sequencing, βA chain starts at Gly |
Endotoxin : | < 1.0 eu per 1 μg of the enzyme as determined by the lal |
Purity : | >90%, as determined by SDS-PAGE and visualized by silver stain. |
Storage : | Lyophilized samples are stable for up to six months at -20° C to -70° C. Upon reconstitution, this cytokine, in the presence of a carrier protein, can be stored under sterile conditions at 2 - 8° C for one month or at -20° C to -70° C in a manual defrost freezer for three months without detectable loss of activity. Avoid repeated freeze-thaw cycles. |
Reconstitution : | It is recommended that sterile PBS containing at least 0.1% human serum albumin or bovine serum albumin be added to the vial to prepare a stock solution of no less than 10 μg/mL. |

Ask a questionDespite the distinct differences, there is still functional overlapping of responsiveness among the populations of activin AB-responding cells.
The characterization of activin-responding cells suggests the involvement of somatotropes (cells that produce growth hormone) and lactotropes (cells that produce prolactin) in activin-induced biological events in the pituitary.
Dysregulation of this pathway could potentially impact trophoblast function and contribute to pregnancy-related complications.
The modulation of activins AB could potentially have therapeutic applications for regulating hormone production and fertility in the reproductive system.
Activin AB produces comparable increases in human trophoblast cell invasion.
Activins AB down-regulate StAR expression, which is involved in the transport of cholesterol for progesterone production.
Activins AB stimulate aromatase/estradiol production in human granulosa-lutein cells.
Activin AB-responding cells were characterized based on their responsiveness in cytosolic free calcium ([Ca2+])i) to four hypothalamic releasing hormones: CRH, GHRH, TRH, and GnRH.
While somatotropes and lactotropes are implicated in activin-induced events, there may be other cell types in the pituitary that also respond to activin and play a role in these events.
Activin AB up-regulates N-cadherin expression in a SMAD2/3-SMAD4-dependent manner.
Activins are composed of homo- or heterodimers of inhibin β subunits and play important roles in the reproductive system.
Activins AB down-regulate StAR expression and decrease progesterone production in human granulosa cells.
It suggest that Activin AB may play a role in regulating trophoblast cell invasion, which is crucial for processes such as placental development and implantation.
The up-regulation of trophoblast cell invasion by Activin AB suggests that targeting this pathway could potentially be explored as a therapeutic strategy for trophoblast-related disorders, although further research is necessary to validate this possibility.
The regulatory effects of activins AB on granulosa cell steroidogenesis are likely mediated via an ALK4-mediated SMAD2/SMAD4-dependent pathway.
Activins AB induce aromatase/estradiol production but suppress StAR/progesterone production in human granulosa-lutein cells.
The information provided specifically mentions the involvement of the SMAD2/3-SMAD4 signaling pathway in mediating Activin AB's effects on trophoblast cell invasion. However, other signaling pathways may also contribute to this process and require further investigation.
Activin AB-responding cells are identified based on their specific responsiveness to activin AB, as observed through changes in cytosolic free calcium ([Ca2+])i) levels. This distinction helps differentiate them from other pituitary cell types.
The hypothalamic releasing hormones tested in relation to activin AB-responding cells are corticotropin-releasing hormone (CRH), growth hormone-releasing hormone (GHRH), thyrotropin-releasing hormone (TRH), and gonadotropin-releasing hormone (GnRH).
Yes, there are distinct differences among each population of activin AB-responding cells.
