• Description: The Protein AG-Cys (His-tag) is an engineered variant of immunoglobulin binding protein. As a fusion construct, Protein AG has multiple IgG binding domains, five of which are from Protein A and two from Protein G. In order to ensure the greatest specificity for IgG binding, regions bounded to the cell wall, cell membrane, and the albumin have been removed. Protein AG has a broader binding capacity than Protein A or Protein G. It binds to all IgG subclasses of humans, rabbits, mice, and rats. It also binds to IgG from cows, goats, sheep, horses, guinea pigs, pigs, dogs, and cats. Thus, it is ideal for purifying monoclonal and polyclonal IgG antibodies. Likewise, our recombinant Protein AG-Cys (His-tag) can bind with almost all types of IgG. Most importantly, it has a single cysteine at C-terminus for direct conjugation. Since the coupling is site-specific, it can create a conjugate with an optimized molar ratio. Furthermore, it has an 8 × His-tag for mild purification. Thus, harsh acidic elution of the conjugates is avoided.
