Description : This gene encodes the enzyme which is involved in the early steps of L-serine synthesis in animal cells. L-serine is required for D-serine and other amino acid synthesis. The enzyme requires NAD/NADH as a cofactor and forms homotetramers for activity. Mutations in this gene have been found in a family with congenital microcephaly, psychomotor retardation and other symptoms. Multiple alternatively spliced transcript variants have been found, however the full-length nature of most are not known.

Applications : Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
Usage : The reaction mixture contained 100 mM Tris-HCl, pH 8.8, 400 mM NaCl, 0.2 mM DTT, 2.5 mM 3-phospho-D-glyceric acid (Sigma P8877), 1 mM NAD, and various amount of PHGDH in a final volume of 50 μL. Reactions were initiated with the addition of PHGDH, and the increase in NADH fluorescence (Ex340/Em460 nm) was continuously monitored for 10 min at room temperature.
