Description : This gene encodes a member of the subtilisin-like proprotein convertase family, which includes proteases that process protein and peptide precursors trafficking through regulated or constitutive branches of the secretory pathway. The encoded protein undergoes an autocatalytic processing event with its prosegment in the ER and is constitutively secreted as an inactive protease into the extracellular matrix and trans-Golgi network. It is expressed in liver, intestine and kidney tissues and escorts specific receptors for lysosomal degradation. It plays a role in cholesterol and fatty acid metabolism. Mutations in this gene have been associated with autosomal dominant familial hypercholesterolemia. Alternative splicing results in multiple transcript variants.

PCSK9-DyLight™ 633 SDS-PAGE Analysis :

Lane 1: MW, Lane 2: PCSK9-DyLight™ 633(2 µg), Lane 3: PCSK9-DyLight™ 633(4 µg). The observed MW of these bands is slightly higher due to post-translational modifications.

PCSK9 is taken up by hepatocytes, downregulating LDL uptake. Huh7 hepatocytes were plated in a 96-well plate at 5x103 cells/well and allowed to adhere. PCSK9-DyLight™ 633 was added at the concentrations indicated for 16 hours in low serum media (2% FBS) followed by a four hour incubation with LDL-DyLight™ 633 at a 1:200 dilution. At the end of the experiment, the cells were stained with 5 μM Hoechst 33342 and then washed three times in PBS and resuspended in live cell imaging buffer. On the right, Hoechst-stained nuclei are blue, LDL-DyLight™ 633 is yellow, and PCSK9-DyLight™ 633, which was added at 20 µg/ml, is red.
