Description : Dendritic cell (DC)-specific intercellular adhesion molecule 3 (ICAM-3) grabbing nonintegrin (DC-SIGN), also known as CD209, is a type II transmembrane protein on DCs with a C-type lectin extracellular domain, is capable of binding ICAM-3 on resting T cells in the secondary lymphoid organs, providing the initial contact between these cells during the establishment of cell-mediated immunity. It is not only a pattern recognition receptor but implicated in immunoregulation of DCs. It has important role in mediating DC adhesion, migration, inflammation, activating primary T cell, triggering immune response and participating in immune escape of pathogens and tumors. DC-SIGN also mediates capture and internalization of viral, bacterial, and fungal pathogens by dendritic cells, such as HIV-1, Ebola virus, cytomegalovirus, Dengue virus, and hepatitis C virus. DC-SIGN is unique in that it regulates adhesion processes, such as DC trafficking and T-cell synapse formation, as well as antigen capture. Moreover, even though several C-type lectins have been shown to bind HIV-1, DC-SIGN does not only capture HIV-1 but also protects it in early endosomes allowing HIV-1 transport by DC to lymphoid tissues, where it enhances trans infection of T cells.

Ask a question
Can you discuss the role of CD209 in shaping adaptive immune responses and its impact on immunotherapy strategies? 01/23/2021

CD209 influences adaptive immune responses by modulating dendritic cell-T cell interactions. Understanding its role is critical for optimizing immunotherapy approaches, particularly in vaccine development.

How does CD209 participate in the recognition of glycosylated pathogens, and what are the implications for innate immunity? 11/14/2019

CD209 recognizes glycosylated patterns on pathogens, particularly through interactions with high-mannose structures. This recognition is crucial for initiating innate immune responses.

Can you elaborate on the genetic polymorphisms of CD209 and their association with susceptibility to infectious diseases? 06/09/2019

Genetic polymorphisms in CD209 have been associated with susceptibility to infectious diseases, including tuberculosis and HIV, suggesting a role in modulating host-pathogen interactions.

How does CD209 contribute to the formation of immunological synapses between dendritic cells and T cells, and what molecular events are involved? 02/09/2019

CD209 facilitates immunological synapse formation by mediating adhesion between dendritic cells and T cells. Molecular events involve interactions with ICAM-3 and other adhesion molecules.

How does CD209 contribute to immune responses, particularly in the context of antigen presentation? 06/02/2018

CD209 facilitates antigen capture by dendritic cells and modulates immune responses by mediating the interaction between dendritic cells and T cells during antigen presentation.
