|Product Overview :||Recombinant Human FASLG protein (Pro134-Leu281), fused to His tag at N-terminus, was expressed in human 293 cells (HEK293).|
|Description :||Fas ligand is also known as FasL, CD178, CD95L, or TNFSF6, is a homotrimeric type-II transmembrane protein that belongs to the tumor necrosis factor (TNF) family. Its binding with its receptor induces apoptosis. Fas ligand/receptor interactions play an important role in the regulation of the immune system and the progression of cancer. Mature human Fas Ligand consists of a 179 amino acid (aa) extracellular domain (ECD), a 22 aa transmembrane segment, and a 80 aa cytoplasmic domain. Within the ECD, human Fas Ligand shares 81% and 78% aa sequence identity with mouse and rat Fas Ligand, respectively. Apoptosis triggered by Fas-Fas ligand binding plays a fundamental role in the regulation of the immune system. Its functions include:T-cell homeostasis, cytotoxic T-cell activity, immune privilege, maternal tolerance, tumor counterattack. Defective Fas-mediated apoptosis may lead to oncogenesis as well as drug resistance in existing tumors. Germline mutation of Fas is associated with autoimmune lymphoproliferative syndrome (ALPS), a childhood disorder of apoptosis.|
|Form :||Lyophilized from 0.22 um filtered solution in PBS, pH7.4, 10% trehalose.|
|Molecular Mass :||The protein has a calculated MW of 17.7 kDa. The protein migrates as 25-30 kDa under reducing (R) condition (SDS-PAGE) due to glycosylation.|
|Protein length :||148|
|Endotoxin :||Less than 1.0 EU per ug by the LAL method.|
|Purity :||>95% as determined by SDS-PAGE.|
|Storage :||For long term storage, the product should be stored at lyophilized state at -20 centigrade or lower.
Please avoid repeated freeze-thaw cycles.
This product is stable after storage at:
-20 centigrade to -70 centigrade for 12 months in lyophilized state;
-70 centigrade for 3 months under sterile conditions after reconstitution.
|Reconstitution :||It is recommended that sterile water be added to the vial to prepare a stock solution of 0.2 ug/ul. Centrifuge the vial at 4ºC before opening to recover the entire contents.|
|SDS-PAGE of FASLG-46H :||
Human Fas Ligand, His Tag on SDS-PAGE under reducing (R) condition. The gel was stained overnight with Coomassie Blue. The purity of the protein is greater than 95%.
