Antigen Description : NF-kappa-B inhibitor alpha, also known as Major histocompatibility complex enhancer-binding protein MAD3, I-kappa-B-alpha, NFKBIA and IKBA, is a cytoplasm and nucleus protein which belongs to the NF-kappa-B inhibitor family. NFKBIA contains five ANK repeats. NFKBIA inhibits the activity of dimeric NF-kappa-B/REL complexes by trapping REL dimers in the cytoplasm through masking of their nuclear localization signals. On cellular stimulation by immune and proinflammatory responses, NFKBIA becomes phosphorylated promoting ubiquitination and degradation, enabling the dimeric RELA to tranlocate to the nucleus and activate transcription. Defects in NFKBIA are the cause of ectodermal dysplasia anhidrotic with T-cell immunodeficiency autosomal dominant (ADEDAID). Ectodermal dysplasia defines a heterogeneous group of disorders due to abnormal development of two or more ectodermal structures. ADEDAID is an ectodermal dysplasia associated with decreased production of pro-inflammatory cytokines and certain interferons, rendering patients susceptible to infection.

Dilution : Western blot: This antibody can be used at 1-2 μg/mL with the appropriate secondary reagents to detect Human NFKBIA in WB. Using a DAB detection system, the detection limit for Human NFKBIA is approximately 2 ng/lane under non-reducing conditions and 0.25 ng/lane under reducing conditions.Direct ELISA: This antibody can be used at 0.1-0.2 μg/mL with the appropriate secondary reagents to detect Human NFKBIA. The detection limit for Human NFKBIA is approximately 0.00245 ng/well.
Preparation : This antibody was obtained from a rabbit immunized with purified, human cell-derived, recombinant Human IkB alpha / NFKBIA

Storage : This antibody can be stored at 2-8°C for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20 to -70 °C. Preservative-Free.Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles.
