    Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. This protein is functionally conserved in its capacity to release the vasoactive peptide, Lys-bradykinin, from low molecular weight kininogen. [provided by RefSeq, Jul 2008]

What is KLK1 protein?

KLK1 protein, also known as tissue kallikrein 1, is an enzyme that is encoded by the KLK1 gene. It belongs to the kallikrein-related peptidase family and is primarily expressed in the kidneys, pancreas, and salivary glands. KLK1 is involved in various physiological processes, including regulation of blood pressure and electrolyte balance.

What is the function of KLK1 protein?

The main function of KLK1 is to catalyze the hydrolysis of kininogens to produce active kinins, particularly bradykinin. Bradykinin is a potent vasodilator that plays a role in regulating blood pressure by promoting the relaxation of blood vessels. KLK1 also exhibits anti-inflammatory properties by inhibiting the formation of pro-inflammatory mediators such as prostaglandins and leukotrienes.

KLK1 related signaling pathway

The signaling pathway of KLK1 is not well-studied or understood compared to other more extensively investigated pathways. However, KLK1 has been shown to have several physiological functions, including regulation of blood pressure, electrolyte balance, and blood coagulation.

KLK1 exerts its effects through the activation of the bradykinin receptor (B2 receptor) pathway. When KLK1 interacts with its substrate kininogen, it generates the vasoactive peptide bradykinin. Bradykinin binds to the B2 receptor, leading to the activation of downstream signaling pathways, such as the PI3K/Akt pathway and the nitric oxide/cyclic GMP pathway. These signaling pathways mediate various biological effects, such as vasodilation, inflammation, and tissue repair.

KLK1 Related Diseases

  •  Hypertension: A decrease in KLK1 activity can result in impaired vasodilation and increased blood pressure.
  •  Renal disorders: Altered KLK1 activity may contribute to kidney diseases, such as chronic kidney disease and diabetic nephropathy.
  •  Fibrosis: KLK1 has been shown to have anti-fibrotic effects, so its deficiency can lead to fibrotic disorders, such as pulmonary fibrosis or liver fibrosis.
  •  Hypertension: KLK1 has been investigated as a potential treatment for hypertension due to its ability to lower blood pressure. It is being studied as an alternative or adjunct to conventional antihypertensive drugs.
  •  Acute pancreatitis: KLK1 has been explored as a therapeutic agent for acute pancreatitis, an inflammatory condition of the pancreas. It has been shown to reduce inflammation, oxidative stress, and tissue damage in experimental models.

Biomedical Application of KLK1 Protein

  •  Hypertension: KLK1 has been investigated as a potential treatment for hypertension due to its ability to lower blood pressure. It is being studied as an alternative or adjunct to conventional antihypertensive drugs.
  •  Diabetic nephropathy: KLK1 has shown promising effects in preclinical studies for the treatment of diabetic nephropathy, a common complication of diabetes that leads to kidney damage. It has been shown to improve kidney function and reduce proteinuria, a hallmark feature of diabetic nephropathy.
  •  Acute pancreatitis: KLK1 has been explored as a therapeutic agent for acute pancreatitis, an inflammatory condition of the pancreas. It has been shown to reduce inflammation, oxidative stress, and tissue damage in experimental models.
