Product Overview : | ACETYLCHOLINESTERASE (EC 3.1.1.7, AChE), also known as RBC cholinesterase, is found primarily in the blood and neural synapses. Low serum cholinesterase activity may relate to exposure to insecticides or to one of a number of variant genotypes. AChE catalyzes the hydrolysis of the neurotransmitter acetylcholine into choline and acetic acid, a reaction necessary to allow a cholinergic neuron to return to its resting state after activation. Cholinesterase levels of cells and plasma are used as a guide in establishing safety precautions relative to exposure and contact, as well as a guide in determining the need for workers to be removed from areas of contact with the organic phosphate insecticides. Simple, direct and automation-ready procedures for measuring AChE activity are very desirable. Acetylcholinesterase Assay is based on an improved Ellman method,in which thiocholine produced by the action of acetylcholinesterase forms a yellow color with 5,5’-dithiobis(2-nitrobenzoic acid). The intensity of the product color, measured at 412 nm, is proportionate to the enzyme activity in the sample. |

Drugs that reversibly inhibit acetylcholinesterase could be used as treatments for Alzheimer's disease and myasthenia gravis, among others. Examples include tacrine and donepezil.
AChE inhibitors have shown some benefit for Alzheimer disease. Inhibition of AChE results in a decreased breakdown and subsequent accumulation of acetylcholine. This excess acetylcholine leads to increased stimulation of muscarinic and nicotinic receptors, which provides some therapeutic relief for the memory deficits in AD.
The primary role of AChE is to terminate neuronal transmission and signaling between synapses to prevent ACh dispersal and activation of nearby receptors. AChE is inhibited by organophosphates and is an important component of pesticides and nerve agents.
Sensitivity: 10 U/L
Whole blood and red blood cells (RBC) lysates can be prepared by diluting 1.0 ml of blood or RBC in 19.0 ml of 0.03% Triton X-100 in water. And then diluted in dilutions in Assay Buffer for further experiment.
Studies have shown that ethanol can inhibit the activity of AChE.
Mammalian Cell Lysis Buffer would be the best lysis buffer to use. <1% Chaps also is OK.
