It is a DNA-specific exonuclease that can catalyze the removal of nucleotides from linear single-stranded DNA in the 3' to 5' direction. It can gradually release deoxyribonucleic acid 5' monophosphate, but the terminal dinucleotide will remain intact. Moreover, it has no activity against double-stranded DNA or RNA. |

Usage : |
1. Reaction system:
PCR products: 5 μL
Exonuclease I (E.coli): 0.5 μL (10U)
2. Incubate the mixture at 37 centigrade for 15 minutes.
3. Inactivate the Exonuclease I (E.coli) at 80 centigrade for 15 minutes.
4. The Purified product can then be used for downstream applications. |
Applications : |
1. Remove single-stranded primers in PCR reactions before Sanger DNA sequencing or SNP analysis.
2. Remove single-stranded primers in nested PCR reactions.
3. Remove single-stranded DNA with 3'-hydroxyl termini from nucleic acid mixtures.
4. Enzymatic purification of PCR products. |
