Thiopropyl Agarose

Cat.no: Thio-001A
Product Overview: Thiopropyl Sepharos contains reactive 2-thiopyridyl disulphide groups attached to agarose through a chemically stable ether linkage, which reacts with solutes containing thiol groups under mild conditions to form mixed disulphides. This reaction forms the basis of covalent chromatography and a procedure for reversible immobilization of thiol containing molecules.
Characteristics: 1. separate thiol-containing proteins and peptides from non-thiol-containing proteins and peptides2. in certain cases separate enzymes with active-site thiol groups from denatured enzymes3. store and protect thiol-containing proteins4. regenerate both the coupled biomolecule and the active bed material
Size: 15 g
Matrix: 6% agarose
Functional group: 2-pyridyl disulphide
Ligand concentration: 18–31 μmole 2-pyridyl disulphide/ml drained medium
Coupling chemistry: approx 10–20 mg protein/ml drained medium, e.g. 14 mg ceruloplasmin (Mr 132 000) /ml drained medium
Recommended column height: 5‐20cm
Chemical stability: Stable to all commonly used aqueous buffers and additives like detergents. Azides should be avoided.
Physical stability: Negligible volume variation due to changes in pH or ionic strength.
pH working range: 2-8
pH CIP range: 2-8
