| Reconstitution : | Collagen type VI dissolves very slowly in 0.5 M acetic acid and requires both vigorous mixing and time. Add small amount of 0.5 M acetic acid, pH 2.5 and use Potter homogenizer (or similar) to break up the protein cake into suspension. Add 0.5 M acetic acid, pH 2.5 to yield 0.1 mg/mL concentration and use vigorous stirring for several days. Concentration higher than 0.1 mg/mL in acetic acid requires even longer time. Use 6M urea (or other chaotropic agent) if application allows it. |
