• Description: Endo S2 is an endoglycosidase that specifically cleaves the N-linked oligosaccharides (Nglycans) on the Fc region of IgG, without affecting the core structure or the Fab region.Endo S2 specifically recognizes and cleaves the glycan chains on native IgG Fc domains. It is often used in antibody structure-function studies, antibody-drug conjugate (ADC) production, and analytical applications to confirm glycosylation state.

• Usage Notes: 1. Recommended buffer system: 20 mM PB or Tris buffer (pH 7.0-8.0) 2. For IgG samples, use a 200: 1 protein-to-enzyme ratio. Maintain protein concentration at 1 μg/μL and incubate in a dry bath at 37 centigrade for at least 30 min.
• Specificity: In 10 mM sodium phosphate, 150 mM NaCl, pH 7.4, incubation at 37 centigrade for 30 min, 1 μg rEndo S2 achieves ≥98% core GlcNAc deglycosylation of 500 μg Bevacizumab.
• Unit Definition: One unit achieves ≥98 % deglycosylation of 1 μg human IgG after 30 min incubation at 37 centigrade in pH 7.4 phosphate buffer.
• LC-MS/MS Analysis: 50 μg Secukinumab was reduced and digested with IdeS for 30 min, then deglycosylated with either rEndo S2 or rPNGase F. Specificity of rEndo S2 glycosidase was confirmed by QE HF mass spectrometry.
