Fibroblast growthfactor binding protein (FGFBP1), also known as HBP17, is a secretedglycoprotein that increases the bioavailability of FGFs. Mature FGFBP1 is a34 kDa, 211 amino acids (aa) O-glycosylated protein with five conserved intrachaindisulfide bonds. FGFBP1 contains a heparinbinding domain (aa 110-143) and adistinct FGF binding region (aa 193-243). Mature human FGFBP1 shares 59% and54% aa sequence identity with mouse and rat FGFBP1, respectively. FGFBP1 isexpressed throughout development and in adult squamous epithelium. |

Measuredby its binding ability in a functional ELISA. 1 μg/mL of rhFGFBP1 was mixedwith serially diluted rhFGF basic. Following incubation, the FGFBP-FGF basiccomplex was captured on a Gt x hFGFBP-coated plate. Bound FGF basic wasmeasured using biotinylated Gt x hFGF basic. The concentration of rhFGF basicthat produces 50% of the optimal binding was found to be approximately 5-20ng/mL. |
