Active Recombinant Human LBP, His-tagged

Cat.No. : LBP-152H
Product Overview : The Lipopolysaccharide Binding Protein is produced from human LBP transfected CHO-cells in serum free medium. Before transfection the complete human LBP-cDNA was amplified by PCR and cloned into expression vector p-POL-DHFR. The recombinant Human LBP was purified by his-tag with metal affinity purification with Talon and controlled by SDS page.Attention: His-tag has no protease site and can not be split off.

Description : The protein encoded by this gene is involved in the acute-phase immunologic response to gram-negative bacterial infections. Gram-negative bacteria contain a glycolipid, lipopolysaccharide (LPS), on their outer cell wall. Together with bactericidal permeability-increasing protein (BPI), the encoded protein binds LPS and interacts with the CD14 receptor, probably playing a role in regulating LPS-dependent monocyte responses. Studies in mice suggest that the encoded protein is necessary for the rapid acute-phase response to LPS but not for the clearance of LPS from circulation. This protein is part of a family of structurally and functionally related proteins, including BPI, plasma cholesteryl ester transfer protein (CETP), and phospholipid transfer protein (PLTP).
Source : Chinese Hamster Ovarian Cells (CHO).
Species : Human
Tag : His
Form : Sterile Filtered White lyophilized (freeze-dried) powder. Recombinant Human LBP was lyophilized from a protein solution (0.25 mg/ml) containing phosphate-buffered saline, pH 7.2.
Bio-activity : Up to 0.2 µg/ ml LBP mediates binding of FITC-LPS (0.5µg/ml) to CD14+CHO transfectants (FACS).
Molecular Mass : On SDS-PAGE commassie blue stained gel, the 95% purified recombinant protein shows a band at 58 kDa.
Stability : Lyophilized LBP Human Recombinant although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution LBP should be stored at 4°C between 2-7 days and for future use below -18°C. Please prevent freeze-thaw cycles.
Reconstitution : Every 10µg of recombinant human LBP should be reconstituted using 40µl of sterile H2O. The solution can be than diluted with phosphate-buffered saline or other buffers.

Ask a question
What happens if LPS cannot make close contact with TLR4? 10/17/2022

If LPS's chemical spatial configuration prevents close contact with TLR4, it exhibits an antagonistic effect, inhibiting the cellular response.

What is the role of Toll-like receptor 4 (TLR4) in this process? 04/21/2022

LPS on the LBP-LPS complex, upon close physical contact with TLR4 on cells, induces TLR4 receptor dimerization or multimerization. This activates various transcription factors, leading to the expression of inflammatory mediators and cytokines.

How does the LBP-LPS complex interact with cells lacking mCD14 receptors? 03/21/2021

For cells without membrane CD14 receptors (e.g., endothelial and epithelial cells), a soluble CD14 (sCD14)-LBP-LPS ternary complex forms, activating TLR4 and inducing various biological effects.

What does LBP stand for? 09/17/2018

LBP stands for Lipopolysaccharide-Binding Protein.

What is the main function of LBP? 02/09/2018

LBP has a high affinity for the lipid A portion in the structure of Lipopolysaccharide (LPS) and forms the LPS-LBP complex.

What is the significance of the LPS-LBP complexes in liver cells? 12/02/2016

Liver cells, similar to macrophages, express mCD14 and TLR4. In the absence of CD14 receptors, LPS can still bind to TLR4, but with relatively weaker intensity.

How does the LBP-LPS complex interact with cells? 08/04/2016

The LBP-LPS complex can transfer LPS to membrane CD14 (mCD14) receptors on cells like monocytes, macrophages, and neutrophils.
