Active Recombinant Mouse Interleukin-2, His Tag

Cat.No. : IL2-403M
Product Overview : Interleukin-2 Mouse Recombinant produced in E.Coli is a single, non-glycosylated polypeptide chain containing 170 amino acids (21-169 a.a) and having a molecular mass of 19.5kDa. The mouse IL-2 is fused to a 20 amino acid His tag at N-Terminus and purified by proprietary chromatographic techniques.

Description : IL2 is a secreted cytokine that is important for the proliferation of T and B lymphocytes. The receptor of this cytokine is a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL4) and interleukin 7 (IL7). The expression of this gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a similar gene in mice leads to ulcerative colitis-like disease, which suggests an essential role of this gene in the immune response to antigenic stimuli.
Form : The Mouse IL2 protein (0.5mg/ml) is supplied in Phosphate Buffered Saline (pH7.4) 0.1M NaCl, and 20% glycerol.
Purity : Greater than 90.0% as determined by SDS-PAGE.
Bio-activity : Measured in a cell proliferation assay using CTLL2 mouse cytotoxic T cells. The ED50 for this effect is < 2.5 ng/ml, corresponding to a specific activity of less than 400,000units/mg.
Physical Appearance : Sterile Filtered colorless clear solution.

In vitro Western blot analysis Forty ng of recombinant human MMP-9 with a C-terminal His-tag (Creative BioMart) was incubated for 2.5 h at 37 °C with 104 CFU of the respective bacteria in PBS (20 μl total volume).. The bacteria-containing samples were centrifuged for 10 min at 3000 x g and the supernatants were mixed with 5 μl 5x non-reducing sample buffer (8 % SDS, 40 % glycerol, 0.02 % bromophenol blue, 250 mM TrisHCl, pH 6.8), separated on a 7.5 % SDS-PAGE and transfer to a 0.45 μm nitrocellulose membrane (Amersham, GE Healthcare, NJ, USA) by semi-dry blotting. The bacteria-containing samples were centrifuged for 10 min at 3000 x g and the supernatants were mixed with 5 μl 5x non-reducing sample buffer (8 % SDS, 40 % glycerol, 0.02 % bromophenol blue, 250 mM TrisHCl, pH 6.8), separated on a 7.5 % SDS-PAGE and transfer to a 0.45 μm..

Commercially available recombinant human hemopexin (e.g., R&D Systems, Minneapolis, MN; Creative BioMart, Shirley, NY; Sino Biological, Beijing, China) has the potential complication of a His-tag, which in the absence of information to the contrary may bind heme or may promote oligomerization of the protein.. 38 , 39 Little or no functional evidence is provided by the vendors to validate the structural authenticity of their recombinant protein.38 , 39 Little or no functional evidence is provided by the vendors to validate the structural authenticity of their recombinant protein.
