Description : UBA6 (ubiquitin-like modifier activating enzyme-6, also known as UbE1L2) is a novel ubiquitin activating enzyme that is also capable of activating the ubiquitin-like protein FAT10 (human leukocyte antigen F-associated transcript 10), which, similarly to ubiquitin, may serve as a signal for proteasomal degradation. Only one UBA6 specific E2 conjugating enzyme, known as Use1 (Uba6-specific E2 enzyme 1) or UbE2Z, which is capable of accepting activated FAT10 and ubiquitin, has been identified to date. siRNA-mediated down-regulation of use1 mRNA results in a strong reduction of FAT10 conjugate formation suggesting that Use1 is the major E2 enzyme in the FAT10 conjugation cascade. Interestingly Use1 has been identified as the first known substrate of FAT10 conjugation, as it is efficiently auto-FAT10ylated. Co-expression of FAT10 and the linker protein NUB1L lead to a proteolytic down-regulation of Use1 suggesting that auto-modification with FAT10 serves as a negative auto-regulation of FAT10 conjugation.

Reconstitution : Reconstitute with 50µl sterile distilled water. Further dilutions should be made with medium containing 5% fetal calf serum or a carrier protein.
