CRISPR (clustered regularly interspaced short palindromic repeat) and CRISPR-associated (Cas) proteins constitute the adaptive immune system in bacteria. This system has been redesigned to create an exemplary genome editing tool for RNA-based therapeutics development. Cas12 and Cas13 have collateral nucleotide cleavage activities and are widely used in CRISPR-based diagnostic platforms. BrCas12b from Brevibacillus shows robust trans-cleavage activity at ideal LAMP (loop-mediated isothermal amplification) temperature. The thermal stability of BrCas12b enables one-pot target nucleic detection to minimize carryover contamination. |
