Description : | The de novo synthesis of pyrimidine nucleotides is required for mammalian cells to proliferate. This gene encodes a trifunctional protein which is associated with the enzymatic activities of the first 3 enzymes in the 6-step pathway of pyrimidine biosynthesis: carbamoylphosphate synthetase (CPS II), aspartate transcarbamoylase, and dihydroorotase. This protein is regulated by the mitogen-activated protein kinase (MAPK) cascade, which indicates a direct link between activation of the MAPK cascade and de novo biosynthesis of pyrimidine nucleotides. Alternative splicing results in multiple transcript variants encoding different isoforms. |

Q&As (5)
Ask a questionCAD is allosterically regulated by UTP, which inhibits its activity, and ATP, which activates it. These nucleotides bind to specific sites on CAD and modulate its function, helping to control pyrimidine biosynthesis.
CAD is a multi-domain enzyme that catalyzes three sequential reactions in de novo pyrimidine biosynthesis: carbamoyl phosphate synthesis, aspartate carbamoylation, and dihydroorotate formation.
CAD helps maintain nucleotide balance by ensuring an adequate supply of pyrimidine nucleotides. When the cell needs more pyrimidines, CAD activity increases, and when there is an excess, CAD is allosterically inhibited.
Researchers use techniques like X-ray crystallography, nuclear magnetic resonance (NMR) spectroscopy, and enzyme assays to study CAD's structure and function. These methods provide insights into its bio-molecular chemistry.
CAD is allosterically regulated by UTP and ATP. UTP negatively regulates CAD, while ATP positively regulates it. UTP binds to an allosteric site, promoting the formation of the less active CAD tetramer, whereas ATP stabilizes the more active CAD dimer.
