Description : | This gene encodes a transcription factor that contains four zinc-finger motifs at the C-terminus and a proline/glutamine-rich DNA-binding domain at the N-terminus. It has an essential role in the normal development of the urogenital system, and it is mutated in a small subset of patients with Wilms tumors. This gene exhibits complex tissue-specific and polymorphic imprinting pattern, with biallelic, and monoallelic expression from the maternal and paternal alleles in different tissues. Multiple transcript variants have been described. In several variants, there is evidence for the use of a non-AUG (CUG) translation initiation site upstream of and in-frame with the first AUG. Authors of PMID:7926762 also provide evidence that WT1 mRNA undergoes RNA editing in human and rat, and that this process is tissue-restricted and developmentally regulated. |

Article Snippet:The beads were washed three times with the wash buffer (40 mM Tris-HCl/pH7.5, 300 mM NaCl, 0.2% NP-40, 1 mM DTT, 10% Glycerol) and eluted with the Flag peptide (Sigma), then stored at ?80°C.The beads were washed three times with the wash buffer (40 mM Tris-HCl/pH7.5, 300 mM NaCl, 0.2% NP-40, 1 mM DTT, 10% Glycerol) and eluted with the Flag peptide (Sigma), then stored at ?80°C.. For the pull-down assay, 400 ng purified recombinant Flag-WT1 (Creative BioMart) and 200 ng recombinant His-Flag-TET2 CD were mixed with nickel beads (Qiagen) in the wash buffer described above at 4°C overnight.. The beads were collected by centrifugation at 2,000 rpm for 3 min at 4°C and bound proteins were eluted with 300 mM imidazole and subjected to SDS-PAGE.The beads were collected by centrifugation at 2,000 rpm for 3 min at 4°C and bound proteins were eluted with 300 mM imidazole and subjected to SDS-PAGE.
