Measles virus (MeV) belongs to the Paramyxoviridae family, Morbillivirus genus. The hemagglutinin (H) constitutes the glycoprotein spikes with fusion (F) proteins, protruding from the viral envelope. The function of the H protein is to bind to a cellular receptor on the target cells, including signaling lymphocyte-activation molecule (SLAM) and nectin-4. MeV H is a 617-residue type II integral membrane glycoprotein. The head domain of the MeV H protein comprises of a six-bladed β-propeller fold. The H protein forms a homodimer linked by an interchain disulfide bond between the cysteine residues at position 154. In the homodimer, the two H protein monomers interact with each other in a highly tilted position. The dimer of the H protein is further assembled into a tetrameric structure by forming a dimer of dimers. |
