Description : Tumor necrosis factor alpha (TNFSF2) is a member of the TNF Superfamily. It is produced chiefly by activated macrophages, but it is produced also by a broad variety of cell types including lymphoid cells, mast cells, endothelial cells, cardiac myocytes, adipose tissue, fibroblasts, and neuronal tissue. The primary role of TNF alpha is in the regulation of immune cells. TNF alpha, being an endogenous pyrogen, is able to induce fever, to induce apoptotic cell death, to induce sepsis (through IL-1 & IL-6 production), to induce cachexia, induce inflammation, and to inhibit tumorigenesis and viral replication.

Effect of the systemic administration of methylprednisolone on the lungs of brain-dead donor rats undergoing pulmonary transplantation

Journal: Clinics    PubMed ID: 24519204    Data: 2014/2/1

Authors: Luiz Felipe Lopes Araujo, Arthur Rodrigo Ronconi Holand, Cristiano Feijó Andrade

Article Snippet:After the samples were thawed, a 96-well plate was coated with monoclonal antibodies against TNF-α and IL-1β.After the samples were thawed, a 96-well plate was coated with monoclonal antibodies against TNF-α and IL-1β.. The wells were filled with either 100 μl of homogenized lung (diluted 1∶2), 100 μl of positive or negative controls, or 100 μl of recombinant TNF-α or IL-1β at concentrations established by the manufacturer (Creative BioMart, NY, USA).. Then, 100 μl of polyclonal anti-TNF-α and anti-IL-1β conjugated to peroxidase was added to the wells, and the samples were incubated for 3 h at room temperature. Then, 100 μl of polyclonal anti-TNF-α and anti-IL-1β conjugated to peroxidase was added to the wells, and the samples were incubated for 3 h at room temperature.

TNF-α levels in lung tissue. There was a significant increase in TNF-α levels in the brain death (BD) group when compared with the methylprednisolone at 5 min after brain death (MP5) and methylprednisolone at 60 min after brain death (MP60) groups. Values are expressed as mean ± SE. *p < 0.001.
