EphrinA2, also knownas ELF1, Cek7L and LERK6, is a member of the Ephrin ligand family which bindsmembers of the Eph receptor family. All ligands share a conserved extracellularsequence, which most likely corresponds to the receptor binding domain. This conservedsequence consists of approximately 125 amino acids (aa) and includes four invariantcysteines. The Aclass ligands have a GPI anchor following the conserved sequence.EphrinA2 has been shown to bind EphA2, EphA3, EphA4, EphA5, EphA6, EphA7, andEphA8. The extracellular domains of human and mouse EphrinA2 share 93% aa identity.Only membranebound or Fcclustered ligands are capable of activating the receptorin vitro. While soluble monomeric ligands bind the receptor they do not inducereceptor autophosphorylation and activation. In vivo, the ligands and receptorsdisplay reciprocal expression.It has been found that nearly all receptors andligands are expressed in developing and adult neural tissue. The Eph/Ephrin familiesalso appear to play a role in angiogenesis. |
