Product Overview: Recombinant human TNF alpha protein (rhTNFa) Val77-Leu233 was produced in human HEK293 cells.
Description: This gene encodes a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. This cytokine is mainly secreted by macrophages. It can bind to, and thus functions through its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. This cytokine is involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. This cytokine has been implicated in a variety of diseases, including autoimmune diseases, insulin resistance, and cancer. Knockout studies in mice also suggested the neuroprotective function of this cytokine.

Bio-activity: The bio-activity was determined by its cytotoxcity assy using L929 Cells with 1ug/ml actinomycin D. ED50<25pg/ml, corresponding to a specific activity of >4X10^7 Unit/mg
Molecular Mass: rhTNFa, without any tag, has a calculated MW of 17.4 kDa expressed. The predicted N-terminus is Val77. Protein migrates as 18 kDa in reduced SDS-PAGE. The gel filtration analysis indicates that only HEK293 expressed TNF alpha protein is trimeric form, which has a molecular mass of 54 kDa.
Endotoxin: Less than 1.0 EU per 1 μg of the rhTNFa by the LAL method.
Purity: >98% as determined by SDS-PAGE. All lots are greater than 95% pure.
Storage: Store at -20? in lyophilized state after receipt. For long term storage, upon reconstitution rhTNFa should be aliquot and store at -20? or -80?. Avoid repeated freeze-thaw cycles.
