Recombinant Human LEPR, Fc-tagged

Cat.No.: LEPR-799H
Product Overview: Recombinant Human LEPR /CD295 Protein, With C-Fc Tag (rhLEPR-Fc) (Phe22-Asp839) was produced in human 293 cells (HEK293).
Description: The protein encoded by this gene belongs to the gp130 family of cytokine receptors that are known to stimulate gene transcription via activation of cytosolic STAT proteins. This protein is a receptor for leptin (an adipocyte-specific hormone that regulates body weight), and is involved in the regulation of fat metabolism, as well as in a novel hematopoietic pathway that is required for normal lymphopoiesis. Mutations in this gene have been associated with obesity and pituitary dysfunction. Alternatively spliced transcript variants encoding different isoforms have been described for this gene. It is noteworthy that this gene and LEPROT gene (GeneID:54741) share the same promoter and the first 2 exons, however, encode distinct protein.
Source: HEK293
Species: Human
Tag: Fc
Form: Lyophilized from 0.22 μm filtered solution in PBS, pH 7.4. Normally Mannitol or Trehalose are added as protectants before lyophilization.
Bio-activity: Measured by its ability to inhibit Leptin-dependent proliferation of BaF3 mouse pro-B cells transfected with rhLEPR-Fc. The ED50 for this effect is typically 0.015-0.13 μg/ml in the presence of 3 ng/ml rhLeptin.
Molecular Mass: rhLEPR-Fc, fused with Fc fragment of human IgG1 at the C-terminus and has a calculated MW of 121 kDa expressed. The predicted N-terminus is Phe22. Protein migrates as 150-165 kDa in reduced SDS-PAGE resulting from glycosylation.
Endotoxin: Less than 1.0 EU per μg of the rhLEPR-Fc by the LAL method.
Purity: >95% as determined by SDS-PAGE.
Storage: Avoid repeated freeze-thaw cycles. No activity loss was observed after storage at: In lyophilized state for 1 year (4°C-8°C); After reconstitution under sterile conditions for 1 month (4°C-8°C) or 3 months (-20°C to -70°C).
Reconstitution: See Certificate of Analysis for details of reconstitution instruction and specific concentration.
