Ask a questionPAK1 has been implicated in cancer progression due to its roles in cell survival, motility, and angiogenesis. Its overexpression has been observed in various tumors and is often associated with increased invasiveness and poor prognosis.
Given PAK1's involvement in cancer progression and other diseases, it has emerged as a potential therapeutic target. Several PAK1 inhibitors are under investigation for their potential in cancer therapy and other conditions where PAK1 signaling is dysregulated.
Once activated, PAK1 phosphorylates a variety of substrates, including kinases, cytoskeletal proteins, and transcription factors. Some of its known targets include Raf-1, MEK1, and LIM kinases.
PAK1 is a serine/threonine kinase that plays a crucial role in multiple cellular processes, including cytoskeletal reorganization, cell motility, and cell survival, primarily through its role in various signaling pathways.
PAK1 regulates cytoskeletal dynamics by phosphorylating and modulating the activity of several cytoskeletal proteins, including filamin, LIM kinases, and myosin light chain, thereby influencing cell shape, motility, and adhesion.
PAK1 is activated through binding to the GTP-bound forms of Cdc42 or Rac, small Rho GTPases. This binding induces a conformational change in PAK1, leading to its autophosphorylation and activation.
While mutations in PAK1 are not commonly associated with hereditary diseases, its overexpression or hyperactivation has been linked to various cancers, promoting cell proliferation, survival, and migration.
