LPS on the LBP-LPS complex, upon close physical contact with TLR4 on cells, induces TLR4 receptor dimerization or multimerization. This activates various transcription factors, leading to the expression of inflammatory mediators and cytokines.
For cells without membrane CD14 receptors (e.g., endothelial and epithelial cells), a soluble CD14 (sCD14)-LBP-LPS ternary complex forms, activating TLR4 and inducing various biological effects.
LBP stands for Lipopolysaccharide-Binding Protein.
LBP has a high affinity for the lipid A portion in the structure of Lipopolysaccharide (LPS) and forms the LPS-LBP complex.
Liver cells, similar to macrophages, express mCD14 and TLR4. In the absence of CD14 receptors, LPS can still bind to TLR4, but with relatively weaker intensity.
The LBP-LPS complex can transfer LPS to membrane CD14 (mCD14) receptors on cells like monocytes, macrophages, and neutrophils.
