Description : | This gene encodes a member of the beta chemokine receptor family, which is predicted to be a seven transmembrane protein similar to G protein-coupled receptors. This protein is expressed by T cells and macrophages, and is known to be an important co-receptor for macrophage-tropic virus, including HIV, to enter host cells. Defective alleles of this gene have been associated with the HIV infection resistance. The ligands of this receptor include monocyte chemoattractant protein 2 (MCP-2), macrophage inflammatory protein 1 alpha (MIP-1 alpha), macrophage inflammatory protein 1 beta (MIP-1 beta) and regulated on activation normal T expressed and secreted protein (RANTES). Expression of this gene was also detected in a promyeloblastic cell line, suggesting that this protein may play a role in granulocyte lineage proliferation and differentiation. This gene is located at the chemokine receptor gene cluster region. An allelic polymorphism in this gene results in both functional and non-functional alleles; the reference genome represents the functional allele. Two transcript variants encoding the same protein have been found for this gene. |

There are still many unsolved questions and future research directions for CCR5 protein. In immunotherapy, the function of CCR5 can be used for disease treatment, such as affecting the migration and activation of immune cells by regulating the expression or function of CCR5, so as to achieve the purpose of treating inflammatory diseases or autoimmune diseases. In addition, antibodies or antagonists against CCR5 may also be considered to inhibit its binding to ligands to further modulate the immune response.
CCR5 protein can interact with other receptors and signaling pathways. For example, in some cancer cells, CCR5 binds to EGFR (epidermal growth factor receptor), triggering EGFR activation and promoting tumor cell growth and metastasis. In addition, CCR5 is also involved in the regulation of signaling pathways such as MAPK (mitogen-activated protein kinase) and PI3K (phosphatidylinositol kinase).
Gene editing technology such as CRISPR-Cas9 can be used to modify or treat the CCR5 gene. By editing the CCR5 gene, the expression of CCR5 protein can be regulated or its function can be changed. This technique is expected to be used in the future to treat some genetic diseases or cancers associated with CCR5.
One way to use CCR5 protein for disease treatment is to use CCR5 antagonists, which are drugs that prevent CCR5 from binding to ligands. There are already a number of CCR5 antagonists in clinical trials for the treatment of diseases such as AIDS and rheumatoid arthritis.
