Cat. No. : KRT18-464H
Description : Keratin 18 is a type I cytokeratin. It is, together with its filament partner keratin 8, perhaps the most commonly found products of the intermediate filament gene family. They are expressed in single layer epithelial tissues of the body. Mutations in this gene have been linked to cryptogenic cirrhosis. Two transcript variants encoding the same protein have been found for this gene.
Source : Escherichia Coli.
Physical Appearance : Sterile Filtered White lyophilized (freeze-dried) powder.
Formulation : The protein (1mg/ml) was lyophilized after from a sterile solution containing 30mM Tris-HCL pH-8, 9.5M urea, 2mM EDTA and 10mM methylammonium chloride.
Purity : Greater than 95.0% as determined by: (a) Analysis by RP-HPLC. (b) Analysis by SDS-PAGE.
Solubility : It is recommended to reconstitute the lyophilized rHuCK-18 in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
Dimers And Aggregates : Less than 1% as determined by silver stained SDS-PAGE gel analysis.
Endotoxin : Less than 0.1 ng/µg (IEU/µg) of rHuCK-18.
Protein Content : Protein quantitation was carried out by two independent methods: 1. UV spectroscopy at 280 nm. 2. Analysis by RP-HPLC, using a standard solution of CK-18 as a Reference Standard.
Reconstitution To Filaments : Performed by mixing equimolar amounts of cytokeratins of type I and type II at concentrations of approx. 0.5 mg/ml, both dissolved in 9.5 M urea buffer (see above). Protofilaments and filament complexes are obtained by dialyzing the resulting polypeptide solution stepwise to a concentration of 4 M urea and then to low salt condition (50 mM NaCI, 2 mM dithiothreitol, 10 mM Tris-HCI, pH 7.4). For immunization purposes, the solution can be further dialyzed against PBS (phosphate buffered saline, e.g. Dulbecco"s PBS).
Stability : Lyophilized rHuCK-18 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution rHuCK-18 should be stored at 4°Cbetween 2-7 days and for future use below -18°C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.

Ask a question
What is the relationship between KRT18 protein and nail diseases? 10/23/2022

KRT18 protein may be associated with nail diseases, such as brittle nails, white spots, and longitudinal cracks. These diseases can affect the hardness and structure of the nails, causing them to appear brittle, deformed, and painful.

What is the relationship between KRT18 protein and hair color? 06/15/2022

KRT18 protein is not directly related to hair color. Hair color is mainly determined by melanocytes, while the KRT18 protein is mainly involved in the process of keratinization.

How can genetic testing be used to understand the genetic variation of KRT18 protein? 04/01/2022

Genetic testing can be done by analyzing DNA sequences to understand genetic variation in the KRT18 protein. These variants may affect the expression level and function of the KRT18 protein, thereby increasing the risk of certain diseases.

How can lifestyle changes affect the function of KRT18 protein? 01/21/2021

Lifestyle changes, such as eating a healthy diet, exercising moderately, and avoiding UV damage, can affect the function and expression levels of KRT18 protein. These alterations can promote the normal function of keratinocytes, which can help maintain healthy skin.

How can KRT18 protein-related diseases be prevented? 08/15/2020

Prevention of KRT18 protein-related diseases is mainly through maintaining a healthy lifestyle, such as a balanced diet, moderate exercise, maintaining water intake, and avoiding UV damage. In addition, regular skin and oral exams can also help detect potential problems early.

What is the role of KRT18 protein in melanoma? 03/13/2020

In melanoma, the KRT18 protein may play a role in promoting tumor growth and metastasis. This may be related to the proliferation and migration of melanoma cells.
