Human Ficolin2 (fibrinogen/collagenlike;previously called Lficolin or ficolinB) is a member of the ficolin family ofsecreted pattern recognition proteins that participate in the lectin complementactivation pathway. Ficolin2 is expressed in the liver and released into thecirculation. The 35 40 kDa, 313 amino acid (aa) human Ficolin2 contains a 25aa signal sequence, an Nterminal collagen domain and a Cterminal fibrinogenlikedomain that includes a calcium binding site and two potential Nglycosylationsites. The collagen domain mediates trimer formation. Larger homomultimers areformed by disulfide links at the Nterminus, the most prominent of which is a12 subunit oligomer. Ficolin2 binds microbial ligands that contain acetylatedcompounds. Mature human Ficolin2 shares 70%, 72%, 76% and 78% aa identity withmouse, rat, cow and pig Ficolin2, respectively. It shares 84% and 52% aa identitywith human ficolin1 and ficolin3, respectively. Single nucleotide polymorphismsare common in human Ficolin2. Some affect serum concentration, while others canincrease or decrease ligand binding. |
