Recombinant Mouse Frizzled Homolog 8 (Drosophila), Fc-tagged

Cat. No.: Fzd8-593M
Product Overview: RecombinantMouse Fzd8 protein was expressed in Murine myeloma cell line with the Fc partof Human IgG1 Pro100-Lys330 (C-terminus) and Mouse Frizzled-8 (Ala25-Pro173)&(Ala28-Pro173)(N-terminus).
Description: Fzd8 is one of atleast ten seventransmembrane (7TM) glycoprotein of the Frizzled family of Wntreceptors. Frizzled proteins are thought to be G-protein-coupled. Wnt engagement, with low density lipoprotein receptor related proteins LRP-5 or LRP-6 actingas co-receptors stabilizes β-catenin and promotes gene transcription that isimportant in development and tissue maintenance.
Source: Murine myeloma cellline, NS0-derived
Form: Supplied as a 0.2 μmfiltered solution in PBS.
N-terminal Sequence: Ala25 & Ala28
Molecular Weight: 43 kDa
Activity: Measured by itsability to interfere with the axis-inducing activity of Xwnt-8 mRNA in early Xenopusembryos. In a population of embryos, addition of 1 ng of mouse Frizzled8/ FcChimera protein per embryo resulted in 50-100% reduction in the number ofsecondary axes produced by 10-20 pg of Xwnt-8 mRNA. Also measured by itsability to bind biotinylated rmWnt-5a in a functional ELISA with an estimatedKD < 1 nM.
Endotoxin Level: <1.0 EU per 1 μgof the protein by the LAL method.
SDS-PAGE: 59-60 kDa, reducingconditions
Purity: >95%, by SDS-PAGEunder reducing conditions and visualized by silver stain.
Reconstitution: Reconstitute at 250μg/mL in sterile PBS.
Storage: Use amanual defrost freezer and avoid repeated freeze-thaw cycles. 12 months from date of receipt, -20 to -70 °C as supplied. 1month, 2 to 8 °C under sterile conditions after reconstitution.3 months, -20 to-70 °C under sterile conditions after reconstitution.
