Description : Cytotoxic Tlymphocyte 4(CTLA-4,CD152), is a type I transmembrane T cell inhibitory molecule that is a member of the Ig superfamily. Human or mouse CTLA4 cDNA encodes 223 amino acids (aa) including a 35 aa signal sequence, a 126 aa extracellular domain (ECD) with one Ig-like V-type domain, a 21 aa transmembrane (TM) sequence, and a 41 aa cytoplasmic sequence. It is widely expressed with highest levels in lymphoid tissues. CD28 and CTLA-4, together with their ligands, B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD28 and CTLA-4 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. CTLA4 transmits an inhibitory signal to T cells, whereas CD28 transmits a stimulatory signal. Intracellular CTLA4 is also found in regulatory T Cells and may play an important role in their functions. Tcell activation through the Tcell receptor and CD28 leads to increased expression of CTLA4.

Ask a question
Are there ongoing clinical trials exploring CTLA-4 inhibitors in combination with other immunotherapies? 05/16/2020

Yes, several clinical trials are investigating the combination of CTLA-4 inhibitors with PD-1 inhibitors and other immunotherapies to improve the overall effectiveness of cancer treatment.

How does the expression of CTLA-4 vary in different types of immune cells? 10/17/2019

CTLA-4 is expressed on various immune cells, including T cells, regulatory T cells (Tregs), and some B cells, contributing to its diverse immunoregulatory functions.

In what ways can CTLA-4 be utilized in organ transplantation? 01/11/2017

Modulating CTLA-4 may have applications in organ transplantation by suppressing the immune response against transplanted tissues and reducing the risk of rejection.

How does CTLA-4 contribute to the maintenance of immune tolerance? 04/30/2016

CTLA-4 is involved in the suppression of autoreactive T cells, contributing to the prevention of autoimmune responses by maintaining immune tolerance.

How does CTLA-4 blockade differ from PD-1/PD-L1 blockade in cancer immunotherapy? 03/18/2016

While both CTLA-4 and PD-1/PD-L1 inhibitors enhance anti-tumor immune responses, they target different checkpoints in the immune system. CTLA-4 primarily regulates T-cell activation in the early stages, whereas PD-1/PD-L1 acts later in the immune response.
