Source : Human cells
Species : Human
Preparation method : Transfected cells were cultured for 48hrs before collection. The cells were lysed in modified RIPA buffer with cocktail of protease inhibitors. Cell debris was removed by centrifugation and then centrifuged to clarify the lysate. The cell lysate was boiled for 5 minutes in 1 x SDS sample buffer (50 mM Tris-HCl pH 6.8, 12.5% glycerol, 1% sodium dodecylsulfate, 0.01% bromophenol blue) containing 5% b-mercaptoethanol, and lyophilized.
Lysis buffer : Modified RIPA Lysis Buffer: 50 mM Tris-HCl pH 7.4, 150 mM NaCl, 1mM EDTA, 1% Triton X-100, 0.1% SDS, 1% Sodium deoxycholate, 1mM PMSF
Quality control Testing : 12.5% SDS-PAGE Stained with Coomassie Blue
Recommended Usage : 1. Centrifuge the tube for a few seconds and ensure the pellet at the bottom of the tube.2. Re-dissolve the pellet using 200μL pure water and boiled for 2-5 min.3. Store it at -80°C. Recommend to aliquot the cell lysate into smaller quantities for optimal storage. Avoid repeated freeze-thaw cycles.Notes:The lysate is ready to load on SDS-PAGE for Western blot application. If dissociating conditions are required, add reducing agent prior to heating.
Stability : Samples are stable for up to twelve months from date of receipt at -80°C
Storage Buffer : 50 mM Tris-HCl pH 7.4, 150 mM NaCl, 1mM EDTA, 1% Triton X-100, 0.1% SDS, 1% Sodium deoxycholate, 1mM PMSF
Storage Instruction : Lysate samples are stable for 12 months from date of receipt when stored at -80°C. Avoid repeated freeze-thaw cycles. Prior to SDS-PAGE fractionation, boil the lysate for 5 minutes.

Ask a question
How is NRG1 implicated in neural development and synaptic plasticity? 06/09/2022

NRG1 is essential for neural development, influencing neuronal migration, myelination, and synapse formation. It also plays a role in synaptic plasticity, modulating neurotransmission and synaptic strength.

What role does NRG1 play in cardiac development and function? 12/01/2019

In the heart, NRG1 is involved in the regulation of cardiac development, including trabeculation. It also has protective effects on cardiac muscle cells and can influence heart repair mechanisms.

Are there any associations between NRG1 mutations or dysregulation and diseases? 01/15/2019

Dysregulation of NRG1 signaling has been implicated in several diseases, including schizophrenia, multiple sclerosis, and certain types of cancers. Genetic variations in the NRG1 gene have been associated with increased risk for some of these conditions.

What are the therapeutic implications of targeting NRG1 in various diseases? 09/18/2018

Given the diverse roles of NRG1 in various physiological processes, it presents as a potential therapeutic target. For instance, recombinant NRG1 has been explored for heart failure treatment, and its modulation is being studied in the context of neural disorders and cancers.

Are there different isoforms of NRG1, and how do they differ in function? 05/29/2018

Yes, there are multiple isoforms of NRG1, generated through alternative splicing. These isoforms can differ in their tissue distribution, receptor binding, and functional roles.

How does NRG1 interact with its receptor, and what are the downstream signaling pathways activated? 03/17/2018

NRG1 primarily interacts with the ErbB family of tyrosine kinase receptors, especially ErbB3 and ErbB4. Upon binding, it activates downstream pathways such as the PI3K/Akt and MAPK pathways.

What is the primary biological function of the NRG1 protein? 02/16/2018

NRG1 is a versatile signaling molecule that plays crucial roles in the development and function of multiple organ systems, including the nervous system, heart, and breast.
