The two plasmids were co-expressed and the IL23 heterodimer was purified.

  2. Measured by its binding ability in a functional ELISA. Immobilized human IL23A-His+IL12B-His at 10 μg/ml (100 μl/well) can bind human IL23R-Fc. The EC50 of human IL23R-Fc is 0.28-0.66 μg/ml.
  3.Measured by its binding ability in a functional ELISA. Immobilized human IL23A-His+IL12B-His at 10 μg/ml (100 μl/well) can bind Cynomolgus IL23R-Fc. The EC50 of Cynomolgus IL23R-Fc is 0.14-0.35 μg/ml.
  4. Measured by its ability to induce IL17 secretion by mouse splenocytes. The ED50 for this effect is 4-20 ng/mL.
• Molecular Mass: The recombinant IL23 heterodimer of human IL23A/IL12B comprises 498 (181+317) amino acids and has a calculated molecular mass of 56.3 (20.1+36.2) kDa. The apparent molecular mass of rhIL23 heterodimer is approximately 22.6 & 42.8 kDa respectively in SDS-PAGE under reducing conditions.

• Stability: Samples are stable for up to twelve months from date of receipt at -20°C to -80°C.
• Storage: Store it under sterile conditions at -20°C to -80°C. It is recommended that the protein be aliquoted for optimal storage. Avoid repeated freeze-thaw cycles.
• Reconstitution: It is recommended that sterile water be added to the vial to prepare a stock solution of 0.2 ug/ul. Centrifuge the vial at 4°C before opening to recover the entire contents.
