Description : Neurofibromin acts as a tumor suppressor and plays a critical role in regulating cellular signaling pathways. The NF1 gene product functions as a negative regulator of the RAS signaling pathway, acting as a GTPase-activating protein (GAP) for RAS proteins. Neurofibromin facilitates the conversion of active, GTP-bound RAS to its inactive, GDP-bound form, thereby modulating cell growth and proliferation. Loss in functional neurofibromin results in dysregulated RAS signaling, leading to abnormal cellular growth and proliferation. This contributes to the development of neurofibromas – benign tumors that arise from peripheral nerves. Dysregulation of RAS signaling pathways can impact various other cellular processes.
