Description : This gene encodes a component of the complement system, a part of the innate immune system that plays an important role in inflammation, host homeostasis, and host defense against pathogens. The encoded preproprotein is proteolytically processed to generate multiple protein products, including the C5 alpha chain, C5 beta chain, C5a anaphylatoxin and C5b. The C5 protein is comprised of the C5 alpha and beta chains, which are linked by a disulfide bridge. Cleavage of the alpha chain by a convertase enzyme results in the formation of the C5a anaphylatoxin, which possesses potent spasmogenic and chemotactic activity, and the C5b macromolecular cleavage product, a subunit of the membrane attack complex (MAC). Mutations in this gene cause complement component 5 deficiency, a disease characterized by recurrent bacterial infections. Alternative splicing results in multiple transcript variants. Ask a question
Can C5 inhibition be a potential therapy for neuroinflammatory conditions such as multiple sclerosis? 01/10/2023

Yes, C5 inhibition has shown promise in treating multiple sclerosis by modulating the inflammatory response and reducing damage to the central nervous system.

How is the C5 protein targeted in therapeutic interventions, and what are the available modalities? 08/25/2022

C5 can be targeted using monoclonal antibodies, such as eculizumab, which inhibits its activity. This approach has been successful in managing various complement-related disorders.

How is the C5 protein involved in the pathogenesis of diseases like PNH? 02/21/2019

In PNH, a mutation leads to uncontrolled complement activation, resulting in the destruction of red blood cells. Inhibiting C5 can prevent this process and improve clinical outcomes.

How does C5 inhibition impact the risk of infections in patients undergoing treatment? 11/16/2016

C5 inhibition can increase susceptibility to certain infections. Close monitoring and appropriate preventive measures are essential to manage the infection risk in patients receiving C5-targeted therapies.

What role does C5 play in the progression of inflammatory diseases like rheumatoid arthritis? 09/01/2016

In rheumatoid arthritis, C5 contributes to inflammation and joint damage. Targeting C5 in treatment may help mitigate the inflammatory response.
