| Description : | LeuDH can catalyze the reductive ammoniation of 4-methylbutanoic acid (salt) to L-leucine or the oxidation of leucine to 4-methylbutanoic acid. LeuDH can also catalyze the reductive amination of α-keto acids with similar structure to prepare corresponding amino acid derivatives. In the process of catalysis, NADH is necessary as hydrogen transporter. |
| Instructions and Precautions : | The following reaction system could be used for reference: 1-10 mg/mL Substrate, 10 mg/mL Enzyme, 10% (v/v) Organic cosolvent, 100 mM Phosphate buffer (pH 7.0), 0.2 mg/mL NAD+, 100 mM Ammonium formate, 10 mg/mL Formate dehydrogenase (FDH). Incubate the reactions at a constant temperature shaker (for example, 30 centigrade, 150 rpm) in for 24~48 hours. Centrifuge each mixture to precipitate protein, them the supernatant can be used for TLC, HPLC or GC analysis. Note: Organic solvent (such as DMSO, methanol, acetonitrile, IPA, etc.) is optional, used for insoluble substrates. |
